A Tris buffer at pH 6.0 (here "Tris" is a nickname use for Tris hydroxymethyl amino methane). For the given pKA table is Tris a compound you would select for a buffer at pH 6.0?

Provided the compound for this buffer, determine the equilibrium concentrations you'd suppose for the conjugate acid and conjugate base species into the buffer at pH 6.0. Suppose that you are dealing along with 1.00 liters of buffer at a complete buffer concentration of 10.0 mM.

Ans:

  • By using carbonic acid, H2CO3:

[A-] + [HA] = 0.010 M

1.44668[HA] = 0.010 M

[HA] = [H2CO3] = 6.9X 10-3M and [A-] = [HCO3-] = 3.1 X 10-3M

  • For MES:

[A-] + [HA] = 0.010 M

1.8128[HA] = 0.010

[HA] = [HMES] = 5.5 Mm

[A-] = [MES-] = 4.5mM
