Question: Assume that you have a 5000 bp double-stranded linear DNA molecule with a gap in one strand 500 nucleotides long, terminated with a 3'-OH group and a 5'-P group. If a DNA polymerase is added to this molecule in vitro (along with everything else needed to synthesize DNA), will the DNA filling the gap be a single piece or consist of short fragments? Would you expect short fragments if the gap was filled in vivo?
