Culture: 24 hours bacterial culture of E. coli and Bacillus on nutrient agar or in broth.

Reagents: Methylene blue or crystal violet

Equipments: Microscopic slides, inoculating loop, bunsen burner, staining tray, and Glassware microscope, lens paper, oil, permanent glass marker, blotting paper.
