Question

Well-developed methods can purify total RNA from cells. After reverse transcribing this RNA into DNA using purified enzymes, the resulting dNA can then be subjected to next-generation sequencing. To assess the protein coding transcriptome, investigators typically purify specifically the mRNA fraction prior to sequencing.

What differences between nascent mRNA (mRNA that is in the process of being transcribed, but has not yet been terminated) and mature mRNA might you use to purify nascent mRNA from total mRNA? Why might adding a step to fractionate the nucleus from cytoplasm prior to RNA purification further improve your results?
