Endospore Lab - Differential Staining for Visualization of Bacterial Cell Structures
SLO 2 - Demonstrate skills of bacterial staining
SLO 4 - Interpret results from selective and differential media
SLO 5 - Demonstrate appropriate use of diagnostic reagents
SLO 6 - Interpret results of diagnostic assays
Purpose: You will determine this. Why are you performing this lab?
Hypothesis: You will come up with this (you are predicting what your results will be)
Introduction Questions:
- What is an endospore/spore?
- What is a vegetative cell?
- What conditions are spores resistant to?
- Name 1 aerobic and 1 anaerobic genera (other thanDesfulfotomaculum) that can exist as vegetative cells or spores.
- Summarize the figure below.
- What is the primary purpose of sporogenesis/germination?
- Summarize the spore staining process (with the following questions)
a. What is used for the primary stain?
b. What cells will become stained - spores/vegetative/both)
c. Why must heat be added during the primary stain?
d. What is used as the decolorizing agent? What does it do?
e. What cells will become colorless? Why?
f. What is used for the counterstain?
g. Which cells will be stained by the counterstain?
h. At the end of the staining process, what is the color of each cell type?
