You may have heard in the news currently that a new species of frog was discovered within a rainforest.

You objective are to isolate genomic DNA and construct an expression library by using λ phage. You are interested within cloning the gene which encodes an enzyme that produces an intense green skin color and also you have an antibody which you think will work.

Questions:

a) Design a well controlled experiment to illustrate that your antibody will work for the library screening. Sketch a picture of your expected experimental effects as part of your reply.

b) Propose two reasonable explanations why you may not be capable to isolate the DNA even if you plated three genomes worth of DNA into your screening.
