Question

Your colleague has now successfully constructed her vector for constitutive (un-regulated) expression of His-tagged HGH, and now wants to proceed with expression of the protein. Unfortunately, it appears that the protein is toxic to the growing bacteria, so she cannot grow enough bacteria to express sufficient amounts of the protein. She and her (incompetent) supervisor are very stressed, but you tell them there is a way to solve the problem, although it would require re-cloning the insert in another type of bacterial expression vector, and also require a small molecule called IPTG.
