Lab - Giant DNA

1) Make up a DNA sequence that is 8 nucleotides long. It can be any sequence you like, but it must include at least one of each nucleotide (that is, 5'-AAAAAAAA-3' would not be acceptable). Indicate the 5' and 3' ends of your sequence.

2) Copy over your DNA sequence from part (1) in the space below. Using that sequence as the top strand, write the sequence of the bottom strand of DNA that would pair with it. Indicate the base-pairs with vertical lines (|) and indicate the 5' and 3' ends of both strands.

3) Suppose that the double-stranded DNA molecule from part (2) were transcribed from left to right. Give the sequence of the mRNA that would be produced by this transcription. Indicate the 5' and 3' ends as appropriate.

Lab - GFP II: Transformation

1) Go onto the course SPOC and watch the Lab Video called "Yeast in the Lab" - it's in the session called "Genes are DNA". Although this is about yeast, it turns out that the behavior of the bacterium E. coli that we will be using is very similar. Answer these questions based on the video; don't worry about the discussion of replica plating. At about 3:09, we see a petri plate with some white stuff on it.

a) What are the white circular spots on the plate called?

b) What are these white circular spots made of?

c) Where did these white circular spots come from?

2) The lab manual talks a lot about pGLO.

a) What is pGLO made of? DNA? Protein? Something else? State the type(s) of molecules and explain your reasoning.

b) Would you expect molecules of pGLO to fluoresce in ultraviolet light? State "yes" or "no" and explain your reasoning.

3) On the back of this sheet, give 3 to 5 short (~1 sentence each) bullet points describing, in your own words, what you will be doing in the lab today. There are a wide range of full-credit answers here; we will grade this as a 1-point check off.

Lab - Aipotu II.

1) Give one hypothesis about the relationship between protein sequence and protein color that you tested in the Aipotu I: Genetics & Biochemistry lab.

2) Give the genotype of each of the four starting strains of Aipotian flowers that you determined in the last Aipotu lab session:

  • Green 1
  • Green 2
  • Red
  • White

3) Briefly describe a strategy that you would employ to produce a pure-breeding purple flower using Aipotu. This need not necessarily work and you do not need to give specific DNA changes, but it must be reasonable.

4) Since today's lab has many parts, this question will be worth 4 points. On the back of this sheet, give 6 to 10 short (~1 sentence each) bullet points describing, in your own words, what you will be doing in the lab today. There are a wide range of full-credit answers here; we will grade this as a 4-point check off.

Lab - PTC II: Wrap-up

1) Give the fragment lengths that you would expect to result from the PCR and restriction enzyme digestion described in the lab manual for individuals with each of the following genotypes. Also, give their phenotype as "Taster" or "non-Taster".

Given:

Allele           Contribution to phenotype

 B                Taster (dominant)

 b                non-Taster (recessive)

Genotype           Bands Present                   Phenotype

 BB

 Bb

 bb

2) What DNA sequence is recognized by the HaeIII enzyme?

3) In the space below, give 3 to 5 short (~1 sentence each) bullet points describing, in your own words, what you will be doing in the lab today. There are a wide range of full-credit answers here; we will grade this as a 1-point check off.
