1. Transfect human tissue culture cells with a reporter gene and plasmids to express either WT or truncated CTD versions of the -amanitin resistant version of the large subunit of RNA polymerase II.

2. Wait 24 hours before adding -amanitin. Add RNA to a GST-eIF4e column. Quantify RNA from the reporter gene that stuck to the GST-eIF4e column vs RNA that flowed through the column. Wait a further 24-48 hours before extracting RNA from cells.
