Question

Bacterial DNA has histone-like proteins, and so a similar protocol to ChIP can be used to study protein-DNA interactions in vivo in E. coli. You decide to study protein-DNA interactions in the lac operon of E. coli under different conditions. You use a crosslinker, an antibody that specifically binds the Lac Repressor protein and PCR primers to amplify the entire lac operon regulatory region.

Name one control you would use in this ChIP experiment and why it is important. You decide to make a reporter gene with 4 binding sites for a new transcription factor ZX2. Draw a simple schematic of the reporter gene below showing the 4 ZX2 binding sites, the reporter gene, and any other elements that are important to include. Include the name of the reporter gene that you choose.
