Discuss each of the following prompts:

-Imagine you use your sterile inoculation loop to take a sample from a chopped up tumor from a carrot. Would you expect a gram stain of that tumor (gall) material to show only one type of microbe, A. tumefaciens? Why or why not?

-Imagine that carrot tumors contain multiple microbes. If your job was to figure out which microbe caused the tumors, how would you get started?

-How would you isolate individual microbes?

-Once you had individual microbes, how would you tell them apart from each other? What lab techniques (there are a few) could you apply to differentiate microbes?

-It takes weeks for tumors to grow, so you would want to narrow down your microbes to pick the most likely candidates, rather than spending months waiting to see whether lots of microbes can cause tumors. If you have a streak plate with isolated microbial colonies from a tumor sample, and you have some tumors to examine, what experiments could you perform to guess which microbes on the plate were most likely associated with the disease, before you go to the trouble of infecting a healthy organism with them?
