Question: The Identification of Unknown Plate Cultures

You're a junior biologist starting your first week of work in a microculture lab. You barely have time to settle into your new job before your boss drops two culture plates on your desk. She asks you to perform some analyses with these two samples: Bacillus megaterium and Escherichia coli. You nervously accept the task, but once your boss walks away you realize you failed to label the culture plates - oh no! You cannot remember which plate is B. megaterium and which is E. coli, so you will first have to perform an experiment to determine their identity. You do not have a microscope available, but you do have the following supplies to help distinguish the unknown cultures:

Gram stain

Two growth media: (1) Phenylethyl alcohol agar (PEA) plates, a growth medium that only allows gram-positive bacteria to grow, and (2) Eosin methylene blue (EMB) agar plates, a medium that only allows gram-negative bacteria to grow
