Question: If you didn't have growth media plates available, what is one other method you could use to assess how the mutation affected E.coli growth?

In this method, a sample of the mutant E. coli strain is inoculated into a liquid growth medium, such as Luria broth, and the mixture is then incubated in conditions that are favourable to growth. Since turbidity measurements display the cell population and growth rate, they can be used to periodically check on the status of the culture. To perform this, either measure the culture's optical density with a spectrophotometer or visually check the culture for changes in cloudiness. This can be done by regularly sampling the culture and comparing its turbidity to that of a control culture. By contrasting the growth rate and turbidity of the modified strain with those of the wild-type strain, one can determine the effect that the mutation has on E. coli growth.
