Questions

1. How would you make a 5X SB buffer from a 28X SB buffer for a total volume of 500mL? Show your solution.

2. From the question above, how much volume of water should you add to your calculated volume of SB Buffer to achieve the final volume mentioned?

3. You were assigned to teach a class of freshmen about the proper ways to handle, use, and maintain a micropipette. How would you go about doing this task? Answer in bullet form.

4. As you were monitoring the work of freshmen students, you noticed that one of them, Tom Riddle, was pipetting volumes of liquid incorrectly. You observed that before the student drew up liquid into the tip of the pipet, there were bubbles present inside the microcentrifuge tube but none on the tip. What caused this error?

5. You corrected Tom, and so he told you that he learned from his mistake. Giving him a second chance, he showed you a second demonstration. You then noticed that there were bubbles present inside the pipet tip this time after drawing up volume from the tube. What caused this error?

6. Having done two errors in a row, you warned Tom that a 3rd one would make him fail the lab activity. And so, Tom proceeded to demonstrate for a third time nervously. This time, no bubbles were present in both the tube and pipet tip. However, you noticed something wrong again. After dispensing "supposedly" the same volume of liquid into three tubes, you observed that the 1st tube had a significantly higher amount of volume than the others, the 2nd tube had lesser volume than the others, and the 3rd tube had the right amount. You made sure that Tom did not adjust the volume knob of the micropipette during the experiment. What could have been the cause of error for tubes 1 and 2?

7. Tom was given a second chance for his 3rd error. He then proceeded with his 4th and final demonstration. He had already drawn up liquid from the stock solution, but the tip of the pipet accidentally hit the outside wall of the tube when he was about to dispense the load. What should Tom do?

8. Before use, all available pipet tips must be ___________.
