Question

Galagan et al (Nature 2013) used a DNA/protein binding approach to map the binding sites for 50 transcription factors (TFs) across the entire genome of the pathogenic bacterium Mycobacterium tuberculosis. Below is Figure 1f from their paper, which shows a histogram of the positions of mapped TF binding sites relative to the predicted start site (first codon - the ATG) of each gene. The authors noted that abundance of TF binding sites in the indicated region (marked with an asterisk) of the histogram was an important and unusual finding from their work.

1. Given what we've discussed, why do you think these binding sites were unexpected, and why are they less frequent than the binding sites located just to the left of the bracketed region of the histogram (in light blue)?

2. Assuming the result is correct, what type of functions might those protein/DNA binding interactions in the indicated region play in regulation within M. tuberculosis?
