The ribosomal cDNA (rcDNA) PCR reactions are a control for the quality of your mRNA and cDNA: any difference between the control and heat-shocked rcDNA PCR reactions is due to experimental error and not due to heat shock regulation, and we have to correct for that error. Estimate the quantitative difference in the results for the rcDNA PCR reactions. For example, if the rcDNA PCR band from the heat shock cDNA is ½ as bright as the rcDNA PCR band from the control cDNA, then write 0.5x. If it's twice as bright, write 2x. Use the results of the PCR reaction from the diluted cDNAs if bands are detectable in both reactions. If not, then use the results of the PCR from the undiluted cDNAs.
