Protein phosphorylation is a post-translational modification and is used as a major regulatory mechanism by cells.

1. You are asked to develop a method to separate the amino acid Serine and Phosphorylated-Serine (Phospho-Serine) using TLC (thin layer chromatography). Please briefly outline two strategies that you may use and provide a rationale for them.

2. If a particular serine residue is phosphorylated in a protein, for example, Serine-15 in p53 (a tumor suppressor protein) is often phosphorylated in cells after exposure to a DNA-damage inducing agent. An immunological method may be employed to detect the Serine-15-phosphorylated p53 verse the unphosphorylated p53. What kind of samples you will need to prepare and what will be in lane 1, 2, 3 of your gel? What primary antibodies you will need for this assay? How kind of patterns of the Western Blot results would you expect?
