We were asked to Design an experimental protocol to determine the solute concentration threshold for plasmolysis.

With 2M of nacl using a 20 fold limit. I really don't understand how to go about the calculations or what they mean about deigning a protocol. These were the questions:

Consider the following when designing your protocol:

  • How will you determine when a cell has experienced plasmolysis? How will it differ from the control cell? What specifically will you be looking for?
  • How will you deal with variability - in other words how will you measure if not all cells in a preparation behave in the same way?
  • How long will you allow the cells to sit in the solution before making your observations?
  • How many cell preparations should you observe?
  • What concentrations should you consider? Should you try a few concentrations first in order to get a better idea of what concentration range to focus on?
  • How will you record data?
  • How will you present your results (table or graph)?
