Competency 208.5.4: Enzymology and Catalytic Mechanism - The graduate constructs models of enzymes, demonstrates how enzymes act as a catalyst in a reaction and factors that influence this reaction, and solves enzyme and catalysis problems.

Introduction:

Proteins are essential to life, and the exact way a protein is made will determine its function. In this assessment, you will write an analysis of how protein structure plays a role in normal protein function, how changes in protein structure can lead to disease, and how enzyme structure and function can be exploited for drug treatment. To complete parts B and C of this task, you must refer to the protein, pathway, and enzyme assigned to you using the attached "ZZP1 Tasks Spreadsheet."

You may refer to the attached "Protein Structure through Genetic Diseases" document as an additional resource to assist you with this task.

You will work through the scenario to respond to part A only.

Scenario

Some researchers are working to find a treatment for sickle cell anemia. They have identified a drug that prevents the subunits of hemoglobin from associating with one another, thereby keeping the hemoglobin proteins as monomers and preventing filament formation. You are asked to make a recommendation about the adoption of this drug as a treatment for Part A.

Requirements:

C. Analyze how an assigned enzyme acts in the assigned pathway, including how a hypothetical drug could impact the assigned pathway, by doing the following:

1. Describe the reaction catalyzed by the assigned enzyme.

2. Explain how feedback inhibition of the assigned enzyme would affect each step in the assigned pathway.

3. Explain how a hypothetical drug can be used to increase the amount of the assigned molecule in the assigned pathway.
