Cloning of the DNMT2 gene
DNA methylation is a heritable epigenetic mark, involving the covalent transfer of a methyl group to the C-5 position of the cytosine ring of DNA by DNA methyttransferases (DNMTs). The DNMT2 gene (alias TRDMT1) is one of this family of DNMT genes responsible for the regulation of DNA methylation. DNMT2 specifically encodes a protein responsible for the methylation of aspartic acid transfer RNA.
The DNA sequence of the DNMT2 gene Is provided for you (below), with the start codon, the stop codon and an internal RE site highlighted.
This is a standard format, showing lust the coding strand for the gene. Each line shows 60 nucleotides of the sequence (indicated by the numbers at the left of each line).
Descnbe how you would clone the DNMT2 gene from DNA extracted from human cells, into the p8luescript ptasmid vector. You should include details of all stages of the experiments required. including all associated calculations. and Illustrating the expected sizes of DNA fragments at each stage. Therefore your work should span the complete cloning process, from PCR amplification of the gene sequence through to verification of a successful cloning event (Hint: the gene conveniently contains a Hind Ill restriction enzyme site which is highlighted in yellow).
You should begin your answer by designing the 2 PCR primer sequences to amplify and digest the sequence ready for cloning. You will need to look carefully at the overall gene sequence and the nucleotide counts to do this.
