A set of mice are lethally irradiated and reconstituted with bone marrow hematopoietic stem cells from a six-week-old donor. The lymphocytes derived from donor versus recipient stem cells can be distinguished based on expression of different allelic variants of the cell-surface receptor CD45. When the reconstituted recipient mice were examined 10 weeks after the bone marrow transfer, studies showed that there were no donor-derived dendritic epidermal γδ T cells (DETCs) in the skins of these mice. What is the best explanation for this result? Bone marrow hematopoietic stem cells fail to home to the thymus to reconstitute the T-cell compartment in the recipient mice. The bone marrow hematopoietic stem cells are unable to give rise to γδ DETCs, as these cells develop only from fetal liver progenitors. The adult thymus of the recipient mice reprograms the donor-derived γδ DETCs to become lymphoid-organ resident γδ T cells. γδ DETCs develop in the thymus of the reconstituted mice but are unable to home to the skin. The adult skin of the reconstituted recipient mice no longer has niches available for the newly developed donor-derived γδ DETCs.
