Research Presentation & Annotated Bibliography word doc", is the assignment file. The project is titled as ‘Biochemical analysis of Restriction endonucleases’. It is a short term two months project in which I determined the optimal conditions (temperature and pH) required for the proper working of restriction endonucleases. These are the class of enzymes that are used to make specific cuts on the DNA. For the experiment, first I isolated the restriction enzymes EcoRI and HindIII from microorganism and then purified it. I took lambda DNA as a model DNA for my experiment as it has restriction sites for both EcoRI and HindIII. Then, I optimized the temperature condition for the restriction enzymes by cutting the lambda DNA by these enzymes at different temperature.

The digested DNA was run in Agarose gel. As the number of cuts is specified for the restriction enzymes on lambda DNA, the well with that specific number of bands will provide the optimized temperature for EcoRI and HindIII. For getting the optimized pH, I prepared the restriction buffers for the enzymes at different pH and repeated the same experiment to get the digested bands in Agarose gel. The well having the specific number of bands for the restriction enzymes will tell the optimized pH for EcoRI and HindIII.

This experiment helped me to know more about the DNA machinery and the impact of restriction enzymes on DNA at different temperature and pH conditions. This relates to my final year project in a way as I’m looking to do that project in this field only. This mini experiment helped me to learn more about the molecular biology technologies that are used in the laboratory. On a larger frame, this experiment can be used to analyze how the restriction enzymes behave at different conditions and this characteristic of these enzymes can be used for any future rDNA technology experiments.
