Assume you have isolated three nonsense mutations in a T4 Phage, each of which is unable to reproduce in Wild Type E.coli. You do not know whether these mutations are in the same gene or not. You infect a wild type (nonpermissive)ecoli with to of these phages at a high MOI (culture A). You observe plaque formation collect the lysate and use half of it to infect a nonsense suppressor strain (permissive) at low MOI (culture B) and the second half to infect wild Type Ecoli (nonpermissive) at low MOI (Culture C). Avg. plaque counts for each of these cultures is given for different combination of phage mutants below.
