The role of a malate binding site ina concavity external to the more deeply situated active site has been a major my stery of the fumarase reaction. The malate, within12 Å of the active site, was bound by hydrogen bonds to two main-chain amides and to two basic residues, H129 and R126. Mutation of the His of this so-called B site of Escherichia colifumarase had little effect on the overall initial rate kinetics ofthe enzyme, which has obscured an understanding of the criticalrole of the site. Contrary to the WT enzyme, which is rate-limitedin the recycling of free enzyme isoforms that follows productrelease, the enzyme with both basic residues modified israte-limited in the product release step itself. A loss ofcomplexity in the mutated, but still functional, step is indicatedby a greatly reduced sensitivity of its rate to changes intemperature. Unlike the inhibition by glycerol shown with normalenzyme and attributed to a viscogenic effect on the recycling rate, the product release step of the B-site mutants is accelerated byglycerol, suggestive of a structural effect on the 12-Å spacebetween the A and B sites. It is proposed that the‘‘extra’’ malate represents a stage in thetransfer of substrate and product between the solvent and the‘‘buried’’ active site of the enzyme.
Justify the hypothesis of early microorganisms were chemolithotroph and why phototrophic organisms that came after had pigments that absorbed maximally at UV and IR radiation
Standard solution:Number drops NaOH solution to restore pink color At rest: Number drops NaOH solution to restore pink color Mild exercise: Number drops NaOH solution to restore pink color Vigorous exercise: Number drops NaOH solution to restore p..
In many countries, conservationists are trying to design national park systems so that "islands" of natural area (the big parks) are connected by thin "corridors" of undisturbed habitat. The idea is that this arrangement will allow animals and plants..
If these three genes were all assorting independently, how many genotypic and phenotypic classes would result in the offspring, and in what proportion, suppose simple dominance and recessiveness in each gene pair?
A couple is about to have a child. They are concerned about the child because of two traits caused by genes on the SAME chromosome.
In mid afternoon, they are on it's under surface, each caterpillar hanging by a few legs. Offer a hypothesis to explain this behaviour. How would you test it.
In the movie, Lost in Space, spider-like creatures attacked members of the Robinson family as they explored an abandoned spaceship. Maureen (the mom) scans the creatures and reports that the spiders are silicon-based life-forms.
IN mice genes for fur color and tail length are located on nonhomologous chromosome. Black (B) fur is dominant to brown (b) and short tails (T) are dominant to long What fraction of the progeny of the cross BbTtxBbtt will have black fur and long t..
Upon completion of genome sequencing projects, how do scientists usually go about asking how many genes there are in genome and where they are located.
Explain why do human beings play a unique role both in degradation and the preservation of the biosphere and discuss the importance of the greenhouse effect.
q1. in a type of plant there are two alleles for flower colour that show incomplete dominance. crcr plants have red
An enzyme that follows the MWC (also known as the concerted model) model for allostery has a T/R ratio of 300 in the absence of substrate. Suppose that a mutation were to reverse that ratio(T/R= 1/300= 3.3 x 10^-3).
