1) Turkish coffee has ~0.854 mg caffeine per mL. There are 0.15 L per coffee cup. How much caffeine is there in a regular coffee cup of Turkish coffee?   

 

The caffeine per ml = 0.854 mg

 

0.15L = 0.15 * 1000 = 150 ml.

 

The caffeine content in 0.15L or 150 ml = 150*0.854 = 128.1 mg.

 

The answer is 128.1 mg.

 

2) What are the steps in a DNA extraction?

 

  1. Lysis - cell and the nucleus are broken open to release the DNA
  2. Precipitation - separates DNA from this cellular debris
  3. Purification - DNA is  rinsed with alcohol to remove any remaining unwanted material and cellular debris and the purified DNA is re-dissoved in water.  

 

3) What are the ingredients in a PCR? What is the purpose of each ingredient?

 

  1. Beginning (template) DNA - DNA sequence which you want copied.
  2. DNA Polymerase - usually Taq polymerase - These enzymes are essential for DNA replication and usually work in pairs to create two identical DNA strands from a single original DNA molecule
  3. Deoxynucleotide triphosphates (dNTPs) -  The four different deoxyribonucleotide triphosphates (dNTPs) are adenine (A), guanine (G), cytosine (C), and thymine (T). These are needed to provide the building blocks for DNA replication. DNA polymerase will add each complementary base to the new growing DNA strand, following  A-T and C-G pairings.
  4. Primers -  provide DNA polymerase with pre-existing nucleotides so that it can begin the synthesis, because DNA plymerase cannot start synthesis on its own.
  5. Buffers - provides a suitable chemical environment
